Evening. I emailed one of the manufacturers of Accelerated Wear Testing systems for Artificial Heart Valves today. I wanted a rough guide on how long it takes to in vitro assess a new heart valve. Some very rough figures. Using their system, I think the most commonly used in the industry, the cycle rate for testing is between 10Hz and 40Hz. It depends on the valve.

So 200 million cycles at 10Hz will take 231 days, 200 million cycles at 40Hz will take 57 days. They might want 400 cycle data, if not longer. At least this gives us a time frame within which to expect some results.

“ RUA Structural Heart

The objectives for RUA Structural Heart for the period was to evaluate heart valve leaflet material and compare the performance of 100% polymeric valves with a novel composite developed by the Group. The computational modelling of the composite material at the design stage suggested that its mechanical properties would be ideal for heart valve leaflets and that there should not be a risk of delamination. The team within the Structural Heart business segment has very recently achieved the initial milestones set for the composite material. The Structural Heart team has achieved the initial milestones set for the composite material and we are delighted to report that after 200 million cycles the material shows no signs of delamination and cut edges remain unchanged as a result of flex fatigue. From a performance perspective, the composite material is very thin and flexible and little energy is required to open a valve and once opened does not restrict blood flow with a good EOA (Effective Orifice Area). Testing has also demonstrated that the properties of the composite restrict crack growth even in damaged leaflets.

100% polymeric valves rely in part on the leaflet design to reduce stress and operate within the performance window of the polymer meaning that polymer would not work in all designs. The composite material retains the blood contacting properties of Elast-Eon but is significantly stronger. This creates the opportunity for RUA Structural Heart to become a supplier of heart valve leaflets to other companies to incorporate in current designs.”

Sorry if teaching people to suck eggs, not my intention. The comment about energy efficiency of the valve caught my eye in the RNS. I think this is important. Both the energy to open the valve and that to maintain flow across the valve are low - if the research is correct. Less energy used by the valve means less disruptive / damaging force being experienced by the valve. Which should mean improved longevity of the valve.

I’m not a bioengineer, but do know quite a bit about blood flow in the human body - please don’t take that as arrogance.

Personally I think this it will be very Important if the major manufacturer confirms RUA’s findings.

Quality posts IV. Very useful to know about the 11p placing, though I should have done more DYOR and already picked up on this! Every day’s a school day. It will get very very interesting with a ‘blow the doors off’ RNS. A muted response to a generally okay RNS is also more understandable. Roll on completion of the HV trial.

Therapeutic Index is the relationship between the dose which has an effect with the dose which a side effect / harm. Might be better thought of as a therapeutic range.

The narrower the index or range, the more likely side effects are unless the dose response / plasma level of drug is measured regularly.

Seeing the disproportionate response the sp has to a relatively small number of trades, a stop loss would get very interesting very quickly. The sp would crash through the floor with any mass sell off. Great if you want a cheap buy in price, otherwise very disconcerting to the rest of us still holding.

Evening NickE, is this a possible scenario?:

The initial 400 million cycle test was possibly done on ‘simple’ Elast Eon samples. This produced a dataset that gets put into CAD/CAM plus genuine ability from the young engineer. A proof of concept HV is then designed, redesigned, built, redesigned, rebuilt… you get the picture, till they’re happy with what they have. This is a proof of concept device that interests the majors - enough for them to at least take Elast Eon seriously - if not the entire valve. This work gets the young engineer of the year award. They’re not lying / misleading with the comment re commercialising this.

What we don’t know is if the majors are simply testing Elast Eon (as stated), or if they are potentially interested in the HV RUA have developed as well?

Spent an hour looking for the RUA patent. I found the application number, but couldn’t find the details of the application. D’Oh!! It would be interesting to see if it’s just the polymer or polymer and manufacturing processes to make it HV compatible which is being patented.

RUA Structural Heart is listed as a dormant company at Companies House. Nothing extraordinary or worrying about that. It just shows the development route RUA is taking at present. Let someone else do the heavy lifting.

Not sure there’s any other way to prime T cells and hence NK cells. The inmate immune system is a very important part of cancer response. We all regularly develop gene mutations which could lead to a cancer developing, the immune system recognises this and deals with it. Only when a cancer cell escapes this does a primary cancer develop.

Do you suggest people shouldn’t take a possibly curative individualised vaccine that might save their life?
