Media outlets rely on press releases. That's why stories appear in several places at the same time containing much the same wording, changed only to expand with additional pulled content (for context) or reduced due to column inches available. The media will only cover the Avacta story if Avacta feed them a ready-made story in a press release. All IMHO of course.

If "P1b will seek to compare Dox vs 6000" then that sounds like the Phase 2 part of a Phase 1b/2 trial. Or a Phase 2 trial, if that can be done without doing Phase 1b. First we need to find RP2D (for STS) and, so it seems, MTD.

The RNS we got on 17thJanuary was highly qualitative. The only quantitative information we got was that 6 biopsies had been taken and analysed:

"Analysis of six tumour biopsies confirms the tumour targeting potential of pre|CISIONTM technology"

"... analysis of tumour biopsies obtained from six patients across several cohorts indicates that doxorubicin is being released within the tumour tissue ..."

"A number of tumour biopsies obtained from patients in different cohorts have also been analysed in order to confirm the release of the active chemotherapy, doxorubicin, in the tumour tissue."

Nowhere did it say that all 6 biospies confirmed the presence of doxorubicin. That quantitative information came during the interview with Paul Hill.

Now we hear that quantitative flesh was put on the qualitative bones yesterday for those privileged invitees ahead of making it known to the whole market. That is what galls when it would have been so simple to do a live webcast. The same happened at the business update during the AGM in June that I would have attended - was all set to attend - had it not been for the train strike on that day: those attending got a lot more from it than those who just had a set of slides to go on.

Well one definition of 'material' is significant; important.

In the context of th estck market and share prices, I'd say 'material' is pertinent to effect on share price movements.

If what has been revealed to us from yesterday's disclosures to privileged invitees has shifted the share price then, yes, what was revealed was material new information.

Very poor show by Avacta. It would have been so simple to set up a livestream of the event. Very poor.

This is the ratio of doxorubicin in tumour vs in bloodstream at the same time. Three things about that.

If AVA6000 is truly specific for FAP, all the doxorubicin will have been generated in FAP environments, predominantly in the TME, so doxorubicin in the bloodstream has come from those FAP environments.

Does 'doxorubicin' mean just doxorubicin or are the less cytotoxic metabolites included? All have to be cleared from the body via the bloodstream.

The >100:1 is a snapshot in time. The PK will be needed to follow how the various concentrations of chemicals change over time. Kinetics means the RATES of reactions/changes.

There will be lots of PK to be presented in graph form to show what the full picture is (which I'll have to have explained to me) but I'm very happy with this news to be going on with.

There is none scheduled in the agenda. Maybe there will be time for a few questions at the end of each presentation. But for the ALS-6000-101 trial, I doubt Andrew Saunders is authorised to talk about future plans.

So here's how it would probably go:

Attendee: "You've RNSed every previous dose escalation with details, but the RNS on 17 January only mentioned a recommendation to continue to higher dose cohorts, with no further details. Has that dose escalation started?"
Alan (stepping in): "We can't tell you about that."
A: "Well can you tell us what dose has been recommended?"
Alan: "No."
A: "Why not?"
Alan: "It's secret."

You can tell whether this person is an investor or a trader from the share BBs commented on.

sdon, your point 3 has no relevance.

IF the amount of doxorubicin found in the tumour were to be low (but it isn't because AVA6000 is targeted to the TME) then, because side effects are low and cardiotoxicity is absent, just give it more frequently. Pfizer have approval for weekly doses of 20mg/m² of doxorubicin.

Or do you distrust Alan when he says therapeutically relevant amounts were found in the tumour across a range of dose levels?
