We have seen Parsortix used for many important studies but this one is particularly significant, not just for Angle but for the CTC liquid biopsy sector as a whole. There is now evidence that ctDNA and CTCs captured from the same patient will produce different results. This means:-

1) When making decisions regarding which therapy to prescribe, ctDNA liquid biopsies alone aren't sufficient - a CTC liquid biopsy will be required
2) By comparing the results of ctDNA and CTC biopsies for patients on treatment, physicians may well be able to track the impact of that therapy and make any necessary adjustments. As AN has repeatedly said, the two types of liquid biopsy are complimentary to one another.

Yes, of course the cash is great but more importantly it's a sign that AZN are actively developing RXC006 and it's not just sitting on the shelf. If the drug performs as hoped then more milestones will be triggered.

Really interesting paper from MD Anderson and Fukushima University was published a few days ago in the BMJ's peer reviewed Journal for Immunotherapy of Cancer. The title of the paper - 'Protein citrullination as a source of cancer neoantigens'. Worth noting that MD Anderson is ranked as the top Cancer Centre in the world and in the top 5 for cancer research and they have highlighted the potential of citrullinated peptides as a target for cancer vaccines.

As you'd hope and expect, Lindy Durrant's Moditope research is heavily cited and is included in the final paragraph of their conclusion:-

'Given the current interest in cancer vaccine development and in immunotherapy, our findings point to the importance of citrullination for antigenicity. Interestingly, we identified several citrullinated proteins that have been targeted for vaccine development, including vimentin and a-enolase.12 13 The importance of citrullination of a-enolase in inducing anticancer immunity has been recently demonstrated.13 Our findings suggest broad relevance of citrullination for inducing cancer immunity.'

The GSK deal comes hot on the heels of a $1.36 billion deal between Bristol-Myers Squibb and Agenus for their anti-TIGIT mAb which also was preclinical. There's also been a stream of other TIGIT deals over the last year or so. It's all about the target and TIGIT is the latest must have for big pharma. Huge amounts being paid even though many believe it's setting is in combination with other checkpoint inhibitors and not as a monotherapy. Worth a quick google for TIGIT mania - it's a thing! Perhaps LD should quickly knock up an avidimab anti-TIGIT mAb.

Another research paper from the John Van Geest Cancer Centre and Scancell has been published today in the peer reviewed journal Immunology and Cell Biology. Again it's early preclinical research looking at targeting the HAGE antigen with an ImmunoBody vaccine.:-

'T-cell responses elicited by the ImmunoBody®-HAGE vaccine were superior to peptide immunisation. Moreover, splenocytes from ImmunoBody®-HAGE vaccinated mice stimulated in vitro could recognise HAGE+ tumour cells and the human TNBC cell line MDA-MB-231. More importantly, the growth of implanted B16/HHDII/DR1/HAGE+ cells was significantly delayed by the ImmunoBody®-HAGE vaccine in both prophylactic and experimental metastasis settings. '

I'd forgotten about the new labs in Oxford - looks like they're now recruiting to get them up and running. Interesting to read the job description of the Senior Scientist - sounds like they may be cracking on with modi TCR research.

Korean biotech, Genexine are also developing a DNA Covid vaccine targeting both Spike and Nucleocapsid proteins although theirs is delivered via electroporation. They've just released phase I trial results - just 21 patients but interesting to see the results which were generally positive although there seems to be a question over the duration of raised neutralising antibody levels.

From Scancell's point of view, it's reassuring to see what inclusion of the N protein as a target brings to the table:-

'The company also examined whether the nucleocapsid protein-specific T cell responses induced by GX-19N were also specific to that of Covid-19 virus variants, such as B.1.1.7 (the U.K.), N.1.351 (South Africa), and P.1 (Brazil).

It found that the amino acid sequences identified by T-cell in GX-19N-vaccinated participants were identical with those of Covid-19 variants.'

'“These results imply that the nucleocapsid protein-specific T cell responses induced by GX-19N can be cross-reactive with emerging Covid-19 virus variants,” the company said.

Not ashamed to admit that I was one of those who thought that the Grand Challenge was in the bag, however, this wasn't due to some evangelistic belief in Scancell. I couldn't see how CRUK could possibly pass up on the chance of having such a high profile team of internationally renowned names working on a Grand Challenge project. I've listed the team below. It's easy to forget how impressive it was - not just the big names that we all recognise, but also the calibre of the institutions represented. They may not have won the challenge, but I believe the fact that these experts were prepared to work on a project led by Lindy Durrant says a great deal about how she is regarded by her peers and I still struggle to understand CRUK's decision.
