Damion- totally agree that Sierra ran two good trials and it is good to see that patients are benefitting in Anogenital Cancer. The treatment should obviously continue, but at what stage of a trial do you reach a conclusion on your data and decide next steps? To me the seven months extension could be seen as a delaying mechanism for not giving Sareum its milestone payment of Six million pounds. They need to open up soon on what’s next. No excuses.

This is not a new research document but there is a good explanation of why SRA737 synergies well with PARP inhibitors ( even though I do not understand some or even all of the technical stuff).

‘ In contrast to other CHK1 inhibitors previously tested in our laboratory, SRA 737 only weakly enhanced ERK1/2 activation and when combined with a Parp inhibitors ( Olaparib) unlike previous studies caused CP95 death receptor activation. It is probable that SRA 737 may have a different spectrum of off target receptors compared to other CHK1 inhibitors that in our present studies are beneficial at enhancing the killing potential of SRA737 when combined with PARP inhibitors.‘

Goes some way towards explaining why SRA 737 is the only CHK1 inhibitor that has not been shelved and why subsequent Research recommended Trials of the SRA737/ Parp inhibitor combo in Small Cell Lung Cancer, Ovarian and Prostate Cancer.

Hoping to see some clues from Sierra in its up and coming quarterly report on how it proposes to take these findings forward- but don’t hold your breath. They will not want to give SRA737 back to Cancer Pioneer Fund(CRUK) given the amount spent on Phase 1/2 trials of SRA 737 mono therapy and in combo with Gemcitabane. But they have no funds to to take forward research that is now well over 18 months old. I hope that they will find an on licencee who will give them a price they cannot refuse. But if not this could get bogged down in the US judicial system. Which would mean patients with an unmet need will continue to suffer.

Just a thought - I have been trying to get my head around the reason for the spectacular trading volumes over the last four trading days without there being a major news announcement. OK we had the Lupus data which showed some future potential but for me that does not trigger volumes like we have seen. I’m making it clear that this is pure speculation on my part but does the pattern of trading suggest in any way that Sareum is being targeted for merger or takeover? Stock prices of potential target companies tend to rise well before a merger or acquisition has officially been announced. Let’s see if the volumes and big buys continue over the remainder of the week - today although higher than normal is a considerable reduction on shares traded last Thursday and Friday. Views welcome.

It’s good to see the current excitement and participation of a large number of shareholders and the future looks bright. It’s amazing how sentiment can change so quickly following on from what I would describe as a period of whinging about the performance of the Company Board. It’s pharmaceuticals and that requires a patient outlook as long term holders will know only to well. SDC-01 and SDC-02 are clearly the jewels in the Crown but I still have high hopes that SRA 737 will be a big earner with its potential in combo with Parp etc in Small Cell Lung Cancer, Prostate and Ovarian Cancers and others. So many routes appear to be coming together at the same time that it is quite frightening to think of what the value would be if they are successful. It may be none of them but for us dreamers it’s a very exciting time. Feel confident that we will break one pence this coming week. At last the message about potential seems to have hit home.GLA

The COVID 19 grant would be nice but the big pay day is the sale of SDC-01 and SDC- 02. I think that is the reason for the Large bulk buying.

Expect to see a continuation of the big delayed buys. It just then depends on the day traders and short termers who seem happy to sell around the current price. Amazed they cannot see the longer term value here. But it takes all sorts.

It was great to see this reference to SAR 20347 in the detailed report - ‘ we used SAR 20347, a Jak inhibitor which can inhibit the whole members of the Jak family to block the Jac Stat pathway. Remarkably, proliferation of FAPs ( responsible for fibrosis and fatty infiltration in muscle injury) was inhibited both in viva and in vitro’.

The in depth report makes fererence to SAR 20347 and SAR 20351. Both have a similar inhibitory profile but SAR20351 showed marked improvement in terms of bio-availability and a superior pharmacokinetic profile.SAR 20351 was also tested on arthritic mice and proved very good invigorate efficacy even with low doses.
