Very good, Jak1 and Tyk2 both inhibit IL-6 but need work in tandem. IL-23 leading to differentiation of Th-17. IL-10 required as precursor, interferon alpha,beta and gamma inhibited by Jak 1 and Tyk2 but signal via different STATS. Tumour necrosis factor alpha and granucyte macrophage colony stimulating factor controlled by Tyk2 Jak1. All the important controls there at the very least to down regulate the cytokine storm l believe.
The added advantage of no to very little toxicity issues as we understand so far. Human trials will confirm this.
Can see no reason for 1801 not to be used as a preventative of a cytokine storm if toxicity confirmed to be inline with current expectations.

Aye Mazik, last sentence says it all in my opinion- Licencing deal as soon as it is granted in 3 months- remember they said they need to protect the IP all last year and Dr Reader said 'the world will know in due course'.

The granting of this patent in the US will provide a further layer of protection around SDC-1802 following the granting of patents protecting the molecule in the US and other major markets in recent years. These patents are important to enhancing the value of the programme as part of our discussions with potential partners
Wow- massive news chaps. Already worldwide patented which means Dr Reader has something special lined up for it. Remember 1802 is orally administered, has no tox, no mtd, got worldwide patent protection as of Jan, no competition in the Tyk2 cancer space and was successfully studied in Lupus by the US Army and Lupus is remarkably similar to covid.
There's that IL-6 which is important in covid-
Specifically, it was reported that treatment with TYK2/JAK1 inhibitor SAR-20351 (now known as SDC-1802) reduces autoantibodies (biological markers of lupus severity) in a spontaneous mouse model of SLE. The data also provided evidence that SAR-20351 inhibits cytokines that play a critical role in lupus, including interferon-alpha, IL-6 and IL-23. The inhibition of IL-23 signalling by SAR-20351 may play a role in the decrease of autoantibodies in the lupus mouse model, as IL-23 signalling drives the differentiation of Th17 cells, which leads to autoantibody production and are pathogenic in lupus.
And with the HNWI's bankrolling us we have no issues of taking it into ph1 which means any licencing deal will double.
