Amendment - approved 29/04/2020;

The SG016 study protocol currently allows patients to take daily doses of the study medication (SNG001 or placebo) at home or in hospital. Hospitalised patients in SG016 who have subsequently been discharged home during the 14-day treatment period, have been successfully taking the study medication at home. SNG001 has also been taken daily at home by patients (with asthma and COPD) in three previous clinical trials with no complications. The current trial allows patients to start treatment as out patients, however we had no workable method to confirm virus positivity or to start patients on their study medication without asking the patient to visit a clinic.

Hospitalised patients treated so far in SG016 have typically had symptoms of COVID-19 for an average of 10-11 days prior to admission to hospital. We believe that it is important to assess SNG001 early in the infection cycle with a view to development of severe Lower Respiratory Tract symptoms and hospitalisation.

This substantial amendment is being submitted to allow patients to be dosed in the home environment within 72 hours of symptoms first developing with either SNG001 or placebo daily for 14 days. The Pilot phase of SG016 now includes 100 patients initiated in hospital and 120 patients initiated in the home setting.

• enhanced antiviral activity in the lungs (measured in sputum and blood samples)
