True 20 to 30 million up.front is pocket money to the big boys.
However it is the smaller pharma that develops up to late stage that will be of interest. They don't have multi billions but will be more than able to.develop.into a commercial.viability
The big pharma more inclined to go for a compound such as 1801 and especially 1802 on safety, pk results and pointers to indications of probable success.

On good results 2 to 4 billion is not out of the question for 1801 and 1802 albeit still a way to go.

What does intrigue me is what is the mode of controlling/treating cancer with mainly Tyk2 inhibitor.

It may be along the lines that Tyk2 can off set a satisfactory degree of off target problems created by the other Jakis at high therapeutic doses as Tyk 2 can indirectly regulate jak3 via pseudo kinase domain JH2 ( allosteric). Just my thoughts on this

If the CRO are stating anticipated dosing of 1801 in first patient on June the 4th then we should hear this month of approval. I take it that the CRO submitted Ethics review at same time as registration thus saving at least a couple of weeks at the least.

737 will take a little while as nothing is fast in the pharma world ( exception maybe CTA approvals down under)

I still maintain a 20 to 30 million upfront payment with high backend milestone payments of which Sareum would receive 27.5%.

Sareum have been very diligent with 1801 i just hope the CPF with 737 do the same. if more than one party are involved then will take a little longer. i would guesstimate around mid to late June before we hear of developments on that front.

Of course the real biggy is SDC1802 but that is well, not as far behind 1801 at it used to be and would put an educated guess that clinical trials of 1802 may well go to Australia also. it would make sense.

I still think there will be a combo somewgere along the line of SCD1802 and 737. SDC1802 in product description to counter the resistance of cancers to gemcitabine as well as compatible as a treatment with 737 and a multitude of other inhibitors as either a supportive or the major form of treatment.

I had a feeling of the upmost confidence today. News is on the horizon me thinks. I come home from work and a very pleasant 15% rise.
Sar are wateretght. However, when 3 rd parties are involved I think this no longer the case.

Oz the other side of the world. More inclined to think it to do with 737. The CPF will be more informed than Sar. There will be people in the know. Sar will not release any news as to 737 is now entering formal discussions that for sure.

My money ( and plenty of it is on 737 news. Strange when you look and you have more shares than have been traded on the day.

Trades are not massive but I would predict a gradual rise now until news is released on either front. Stlll seems a little early to me for news on 1801, it will come when ready.

Good morning C79. Since Alopecia is basically an auto immune disease I see very little advantage of a topical dressing.
The same I feel can be said about Psoriasis and indeed one pharma has already discontinued this approach.
Autoimmune is about controlling the production of new cells and and hence see very little benefit to the topical applications.

Tyk 2 through the pseudo kinase domain will regulate JAK2 and 3. Jak2 and 3 have far greater success albeit associated safet concerns in IBD. TYK 2 on its own has not proven favourable in this area.

Hence, we can assume that the addition of an optimum amount especially of Jak1 with selectivity over Jak 2 and Jak3 should prove more favourable. All about getting the balance right.

Early Jak inhibitors were not selective enough. One size fits all clearly has not proven to work in this instance indicated by their off target effects, leading to less than effective therapeutic treatment levels ( due to toxicity) being administered to obtain the desired reponse.

737 had already been valued at over 400 million on the estimation by Edison group. Originally it were 700 million dollars. There was no update from Sareum at this time.

The path forward was given as rapid on licence or develop further then on licence as per AGM.

It is in the hands of the CPF and not Sareum.

6 weeks and no update has little relevance. Negotiations will be on going.

Should we take the second option of carrying out further develoment, then this itself does not rule out an on license.

Are you seriously expecting an RNS from Sareum stating that we have decided to carry out further development and as such during this time no on licence will be available?

Rapid on licence or any licence regarding the pharma industry takes place over months. Yes informal discussions take place as to how much is paid up front, milestones and risk mitiagtion. Then a further period of formal communications will follow which may or may not involve the agreement of an on license.

Any pharma will require data to evaluate and a time period for doing so. As Tim stated in the AGM looking at one day to a month to evaluate genetic profile data dependant on how it has been presented back to the CPF , substantially thick folders.

Now we look at the whole caboodle and you getting impatient as 6 weeks and no update.

If yo were a pharma would you not be looking up time scales and hurdles in maybe applying for clinical trials in areas of preclinical investigation carried out by SO? Pre clinical ie PARPi and PD-1 in combo with LDG?

Government institutions. Just completed my security clearance, all 38 pages. Online of course. I fill in with all information. partners I have had. Names addresses, date of birth and establishments I have worked at.

An example here is address. Name of organisation you work for. I name the organisation, Address name and they want the name of the street. Its a big place but has no street name. You enter the address I have, and when you get to end of form to submit gets rejected as no street name. I put down A308 which is a road 30 miles long. It is accepted of course,
A post code and a large company has no street name I have to be careful here as MOD. posts i have put before comes up with a breach screen.
They will ask previous supervisors names. Contact details. Gets rejected as requires middle names. Past 10 years and over 15 different establishments it gets difficult. they require work and home contact numbers and email addresses. Some of these people now deseaced
Now bare in mind I have also had this clearance for 7 years.
Every detail they want to know.

My expectations as to trial approval and SP at 170 to 180 exact.

Now the 50k pay upfront for Aurora knocked the price up but not due to the 50k upfront but also the 900 k possible milestone payment of 900 k on success of solving solubility issues with regards to the oral route.

CHK1 much development has happened since SO cessation of its progress. It is vital as mutant P53 involved in over 50% of cancer tumours.

Any development in 737 will be worth a multitude of any upfront payment, as potential milestones will follow

Sierra Oncology would have been liable in any future development to pay a 2nd milestone of 12 million dollars. This at the same time that SO were diverting all resources into momo and even reducing the funding the advancement of 737.

I still stick by a minimum upfront payment of absolute minimum of 25 million upfront payment with high back ended milestones. Of course we only get 27.5% as we know but the potential of a revenue stream will propel Sareum SP

1801, once approval granted opens the way for 1802. Should dose escalation studies prove favourable on its own then market cap of a mere 200 million not out of the question irrespective of 737.
Announcement of application of 1802 clinical trial application should as a minimum put a 400 million market cap.

737 on licence, 1801 approval with good phase trials leading to 1802 in clinical trial for projected indications looking circa 800 million.

Downside of course is Sareum cannot rum forever without further funding. Hence any milestone payment form 737 will transform the SP.

Although Sareum objective to on licence late preclinical or early clinical, Many moons ago stated that it would give greater share holder value to on licence at later clinical stage.

A while away from the 4 billion valuations. Bare in ming that the HNWI's have options I think from memory run to 2025 ish

Once the ground is settled and the risk removed from 737 with regards to on licence then a takeover becomes a very viable option. When risk is removed the value increases. Although I feel moons away a 4bn takeover is not out of the question, years later down the line it may well be deemed by some pharma to be a bargain.

Followed by, as we have suffered from the effects of Covid19 and also consequent shortage of experienced staff we are at the moment doing the best we can to address issues you have raised. It may be prudent for companies to submit clinical trials applications to other countries as they are not experiencing the shortfalls of experienced staff and little to no backlog exists. Whilst we strive to be on the cutting edge of approving trials adhering to clinical trial applications adhering to the latest newly updated compliance policies, with the potential to be a world class contributor in the very near future, there are likely to be delays caused by lack of communications between legislative bodies and authorities involved as such, along with newly recruited staff that are undergoing training.

May I suggest you consider taking your companies trial application to a country like Australia which welcomes clinical trials in an endeavour to become a world leader in clinical trial development?
It is a simple but effective process which involves evaluation of your trial by an ethics committee after submitting a clinical trial notification. The end decision is made by the research establishment that will carry out the clinical trial after taking in advice from the ERC. Their processes are streamlined. There is much less red tape.

It is indeed encouraging. Clinical trials need to be approved for SDC 1801. Good safety results will lead to increased value in SDC 1802 too.

On approval with goid toxicity results ought to put up around the billion mark plus. Not much for what we have.
