Tyk2 signals via JH2 domain. All other jaks basically signal via JH1 domain. Jak 2 patially signals both and JH1 and JH2. JH2 best thought of as a remote control switching on or off certain but limited receptors in JH1. Whilst very selective in this process it does not address on off control in the JH1 domain that certain autoimmune disorders seek to be controlled. JH1 domain is enzyme domain ie directly the active site. JH2 is a pseudokinase domain. ie does not directly control the active site.
Did have slight disagreement with Thoth2 over this.
TYK2 is pseudo kinase domain allosteric. Jak1 2 and 3 are JH domain 1.
Therefore with our inhibitor we not only have control of the allosteric site with the TYK2 but also the non allosteric site with Jaks 1 2 and 3.

We are not pure Jak1 but have at least 5 five fold selectivity of Jak 1 over Jak2 and Jak3.

Pure Tyk 2 will have its limitations as is coming to light with the treatment of IBD. These failings already coming to light with BMS proving OK in psoriasis but achieved nothing in IBD.

Good afternoon Bassethd, I concur fully in that it would have been beneficial to update its shareholders even to state that matters in hand are progressing.
In their RNS released
'Sareum and its advisors are seeking additional clarification from the MHRA and will provide an update on next steps for resubmission as soon as possible.'

They would have been better off leaving that out.
Scientists are scientists and do become very focused on their work to the detriment of good communications. I dare say they are addressing the issue with regards to GLP compliance but at the same time not addressing a major issue causing a declining share price and that is uncertainty brought about by lack of an update to reassure investors.

There will be masses of documentation going back to around 5 years relating to development of SDC1801 and this will include SDC1802. May take a while to resolve but feel confident of the end result.

SRA737 formerly CCT245737 a Sareum and CRT developed Checkpoint Kinase 1 inhibitor that is looking increasingly attractive as time goes on. Much work be carried out since 2019.

Excellent results in combo with Niraparib carried out in 2018 ( owned then by Tesaro before being bought out at beginning of 2019 by GSK. Much more carried out since, as indicated by patent protection of SRA737 in combo.

At phase 1 on licence to Pronai Therapeutics ( Sierra Oncology) the agreement was around 300 million in milestones plus royalties of circa 10% of which Sareum would receive 27.5% of.

Good results in combo with LDG has been concluded in clinical trial ( hence patent)
SRA737 far superior to Prexasertib as far more selective over CHK2. ( it was originally believed that CHK2 to be far more effective over and in combination with CHK1 and the new idea at the time circa 2008, however this has not proven to be the case)

For what we have the current SP is peanuts. As for the uncertainty that comes under market sentiment. never quite fathomed that one out. Nearest I got was whichever way you think the SP will move the market sentiment pushes it into the other direction whilst taking no prisoners.

should Sareum have submitted/resubmitted info within the 14day period then there is a 16 day (stated as maximum) period of when the MHRA will give final decision. If they have applied for an extension to this time it will then be 23 days following receipt of submission/resubmission.
Within the 14day period RFI Sareum have either responded with the requisite information or applied for an extension.
It would have been good to receive an RNS detailing this.
RNS should be received within 31 days from the 8th of November which makes your thinking of an RNS this week correct, should time frames and non extension have been applied for.

To make a request, email the MHRA at clintrialhelpline@mhra.gov.uk advising how much time beyond the usual 14 days is needed. If granted, you’ll submit your response to the RFI within the agreed timeframe. A final decision will be issued 10 days after your response has been received. Much been done with 737 including combo with WEE1 last year Krone. Unfortunately the patented SRA737 with LDG would have restricted development in some fields.

It will turn out OK Matty Boy 1965. The war in Ukraine has dragged AiM down over 40%.

All the MHRA have done is requested supporting evidence of GLP. 51% of CTA applications are requested for further information and of these 95% are granted. If they can't provide the prerequisite information now now they never will. Should not prove too troublesome as l dare say the same CRO who are developing SDC1802 now have carried out pre clinical.testing in 1801.

Work of this nature is not given to amateurs. They carry out much testing and data analysis. They are costly to use. Far more chance of getting unmanipulated data here as opposed to a large pharma that have their own labs and KPI's to go along with it.

It is not the molecules as such that are protected but the preparation of. Effectively you would not be able to create a generic drug of the same calibre as the preparation of is now patented. Crytallisation of a compound also comes into play here. Both 1801 and 1802 are protected.

Interesting point Carter with regards to vaccines. If you have covid early stages and then have a vaccine does the Covid then mutate as a resistance to the vaccine?
The vaccine supposedly takes a period of time for you to build up immunity. In effect you are subjecting the virus to a partial resistance.
Viruses can be subject to ultra violet light. In low exposures they will develop resistance to.
Covid will be round for years to come yet.

HbD. I would assume that we are using the same CRO for 1802 as we used for 1801.

CTA approval, whilst not done and dusted is bound to cause worry to some in light of the recent rather poorly worded RNS. I do not see as an issue that cannot be resolved.

No mass sell up of investors of 66 million shares. Clearly not sufficient concern to them to sell up in their droves.
