Sanofi: First patient outside U.S. treated in global Kevzara® (sarilumab) clinical trial program for patients with severe COVID-19

PARIS and TARRYTOWN, N.Y. – March 30, 2020 – The first patient outside of the U.S. has been treated as part of a global clinical program evaluating Kevzara® (sarilumab) in patients hospitalized with severe COVID-19. The global clinical program has now been initiated in Italy, Spain, Germany, France, Canada, Russia and the United States – all countries that have been impacted by COVID-19.

This is the second multi-center, double-blind, Phase 2/3 trial as part of the Kevzara COVID-19 program, and the companies are continuing to work with health authorities around the world to secure initiation at additional sites. This follows Sanofi and Regeneron’s announcement earlier this month of the initiation of the first trial, which is U.S.-based.

 London's Excel Exhibition Centre, which re-opens next week as the NHS Nightingale Hospital Excel ( housing up to 4000 patients ) , will presumably be the most obvious venue for urgent and imminent patient trials...Good luck to everyone!!! :-)

Ps Confirmation that patient trials are underway will see this jump in value significantly imho, and any therapeutic benefit for the treatment of CV reported from the upcoming patient trials will see this multi bag exponentially imho....the current bargain low offers potential tremendous upside. A potential blockbuster for the treatment of CV patients, and so given the global crisis at hand, could already be attracting the attention and interest of Major Pharma's who might want to invest and assist in its development. Gla ;-)

Tiziana plans to administer TZLS-501 using a proprietary formulation technology. The company entered into a world-wide license for composition-of-matter of TZLS-501, a fully human MAb targeting IL-6R, with privately-held Swiss biotech Novimmune in 2017.

Certain patients infected with coronavirus COVID-19 may develop an uncontrolled immune response (cytokine storm) resulting in severe damage to lung tissue which could lead to respiratory failure. Early clinical studies conducted by doctors in China suggest that anti-IL6R MAbs may be used in clinical practice for treatment of COVID-19.

China already recommending Actemra and Kevzara for such lung damage

Consequently, China’s National Health Commission has recommended the use of Roche’s (ROG: SIX) blockbuster drug, Actemra (tocilizumab)for treatment of patients infected with COVID-19, with serious lung damage and elevated IL-6 levels. Actemra was first approved by the Food and Drug Administration in 2010 for rheumatoid arthritis. Besides Actemra, Sanofi and Regeneron are currently exploring Kevzara (sarilumab), an FDA-approved anti-interleukin (IL)-6 receptor therapy for rheumatoid arthritis, for treatment of severe COVID-19.

Tiziana’s anti-IL-6R MAb binds to both the membrane-bound and soluble forms of IL-6R and rapidly depletes circulating levels of IL-6 in the blood. An excessive production of IL-6 is regarded as a key driver of chronic inflammation and is believed to be associated with severe lung damage observed with COVID-19 infections and acute respiratory illness. A recent Chinese study also reported that COVID-19 infection caused clusters of severe respiratory illness such as severe acute respiratory distress syndrome (ARDS).

“We believe that the features of TZLS-501 consisting of its dual mechanism of action to inhibit signaling by the membrane-bound and soluble IL-6 receptors along with rapid depletion of circulating IL-6 cytokine, a major cause of lung damage, provides TZLS-501 with distinct advantages for treatment of COVID-19 over other anti-IL-6R MAbs such as Actemra and Kevzara for treatment of COVID-19. The recent decision by researchers in China to add Actemra to treatment guidelines for coronavirus patients with serious lung damage confirms the utility of anti-IL6R MAb.

CloudCoCo Group plc (AIM: CLCO.L), formerly Adept4 plc ("Adept4"), the AIM quoted provider of IT as a Service is pleased to announce final results for the year ended 30 September 2019 ("FY19"). These results reflect the performance of the legacy Adept 4 business only over what was a well-documented challenging period.


FY19 Summary


· Revenue in the year of £7.3 million (FY18: £10.2 million) and gross profit £3.7 million (FY18: £5.7million), reflecting the reduced emphasis on new business generation after the Board's strategic review;

· Trading overheads2 fell by 23% to £4.0 million (FY18: £5.1 million), due to reduced cost base required by as a result of the Board's decision to focus on existing customer base;

· Resultant Trading Group EBITDA1 loss of £0.2 million (FY18: profit £0.6 million); and

· Reported loss after tax of £5.2 million (FY18: £3.8 million), stated after a significant non-cash impairment charge of £3.0m against goodwill and other intangible assets.


Post Year End


· Completion of acquisition of CloudCoCo Limited on 21 October 2019, bringing a strong and experienced sales and business development team to the Group;

· The acquisition was satisfied by the issue of 218,160,568 ordinary shares of 1 pence each in the share capital of the Company, representing a total value of £7.2 million on completion;

· Appointment of Andy Mills, former Chairman of CloudCoCo Limited as Chief Executive Officer of the Company;

· Refinancing of the Group's debt, reducing loan note debt from £5.0 million to £3.5 million and extension of a new £0.5 million working capital facility;

· Rebrand and change of the Company's name to CloudCoCo Group plc on 29 November 2019;

· New business already being won and value of sales opportunities growing evidencing strong sales pedigree of CloudCoCo Limited team.

Blackstone Group Inc. President Jonathan Gray said the private-equity giant is “bullish on the U.K.” and sees potential for more deals.

“Right here in the U.K. there’s a lot of opportunity,” Gray said in an interview with Bloomberg Television in London on Thursday. “Brexit has obviously added an element of uncertainty and has slowed growth, but the U.K. stock market in dollar terms has underperformed by 50% relative to the U.S. market, and that seems to us to be too much of a discount.”

 The JV theory, is conceivable but not just yet imho....Confirmation of the Credit Facility will get this flying, especially on disclosure of who the Asset Manager is, which will very likely see a flood of new investors including institutional investors on the backing of a leading renowned world wide Asset Manager, as well as a flood of new investors into the Blackoak Alpha Fund with such illustrious backing....and then maybe a JV thereafter.
