Genflow Biosciences Ltd (LSE:GENF, OTCQB:GENFF, FRA:WQ5), a biotechnology company focused on gene therapies for age-related diseases, has entered a technology collaboration with Acuitas Therapeutics, the Canadian lipid nanoparticle (LNP) delivery specialist whose platform underpinned several of the most widely used mRNA Covid-19 vaccines.

Under the agreement, Genflow will provide its proprietary SIRT6 mRNA payload to Acuitas, which will formulate the material using its LNP platform before returning the resulting formulations to Genflow for preclinical evaluation SIRT6 is a protein-coding gene associated with DNA repair and longevity, and Genflow's platform is designed to harness its therapeutic potential in age-related disease.

LNP technology acts as a delivery vehicle for mRNA-based medicines, encapsulating genetic material in fatty particles that protect it from degradation and enable uptake by target cells.

The collaboration is fully funded by Acuitas, with no cash payable by Genflow and no dilution to existing shareholders.

Genflow said it does not expect the arrangement to have a material impact on near-term revenues, but the board regards Acuitas' willingness to commit its own resources as an independent validation of the company's science.

Data generated through the programme are expected to inform decisions on delivery optimisation, target tissue engagement, and the structuring of potential future partnering and licensing arrangements.

The current agreement is focused on initial preclinical activities but establishes a formal framework for potential further strategic engagement between the parties.

Eric Leire, chief executive of Genflow, said the collaboration was one of the most significant steps the company had taken in developing its SIRT6 platform.

"Acuitas are the gold standard in LNP delivery and the fact that they are committing their own resources to work with our science is a powerful endorsement of what we are building," he said

Not too caught up with the day-to-day share price activity, my attention is drawn to see how well the company has done and subsequent share price appreciation by the year end.

Eli Lilly’s latest move could open new frontiers in drug development — and aptly, it’s going through the Gate.

In a deal worth up to $856 million, Lilly has partnered with Gate Bioscience, a California-based biotech pioneering a new small-molecule drug class called “molecular gates.” These engineered molecules aim to selectively eliminate disease-causing extracellular proteins — a category often deemed “undruggable” by conventional means.

A significant proportion of disease-driving proteins operate outside the cell, and many remain inaccessible to traditional small molecules or biologics. Gate’s approach could change that. By designing molecules that act like precision-targeted disposal systems, they may offer oral drugs for conditions like neurodegenerative diseases, inflammation, and cancer — diseases where new modalities are desperately needed.

What makes this collaboration particularly interesting is that it’s part of Lilly’s Catalyze360 initiative — a venture-style model to support breakthrough science at early stages. Gate, still in preclinical development, will receive R&D support from Lilly ExploR&D, benefiting from Lilly’s deep expertise and resources in translational development.

Gate Bioscience only emerged from stealth last year with backing from Versant, a16z Bio+Health, ARCH, and GV. This is their first major public partnership — and it’s a big one.

The “molecular gate” concept exemplifies the convergence of protein engineering, structural biology, and smart chemistry to rethink what small molecules can do. If successful, this could lead to drugs that are both highly selective and orally available — the holy grail in many therapeutic areas.

It’s still early days, but this is a space worth watching.

Patent portfolio strengthened with international patent application under the Patent Cooperation Treaty

Genflow (LSE: GENF), is pleased to announce that it has filed an international patent application in the US under the Patent Cooperation Treaty ("PCT") related to variants of Sirtuin-6 ("SIRT6"), and the gene variant's therapeutic uses in the regulation of aging and for the treatment and prevention of age-related diseases (the "Application").

The patent has been filed jointly with the University of Rochester, a prestigious private research university in New York, and Genflow holds the exclusive licence through Genflow Biosciences SRL (the Company's Belgian subsidiary). The Application claims priority from the provisional application US 63/188,573 of May 14, 2021. Full details of the exclusive licence are set out in the Company's prospectus which can be found on its website.

The Company believes that the invention set out in the Application has the potential to enhance both health-span and, possibly, life expectancy. The SIRT6 gene variant found in centenarians (people aged more than 100 years old) has demonstrated significant capabilities in epigenetic tissue rejuvenation and therefore, constitutes an excellent target to prevent and treat age-related diseases. The ability of SIRT6 to act on the rejuvenation of cells is indicative of the SIRT6 gene mutation's potential to not just halt the aging process, but to reverse it.

The filing will strengthen Genflow's intellectual property portfolio establishing a patent footprint for the Company in the US and internationally. By filing an 'international' patent application with the PCT Genflow is allowed to seek patent protection for the relevant invention simultaneously in a large number of countries. In addition, use of the PCT will reduce administration for the Company and lead to lower costs and moreover, support the Company's decisions concerning the prosecution of the application before different national patent offices in the PCT National Phase of processing.

This Application is in addition to a previously filed patent application for the method of administration and delivery of the Company's product into humans and dogs, using Genflow's lead compound, GF-1002, a recombinant self-complementary adeno-associated virus ("AAV").

Overexpression of Sirt6 reduced tumor volume significantly (40% reduction, n=7, p<0.01) and increased muscle mass by five-fold in the melanoma-challenged male mice, compared to their tumor-bearing control mice (n=10). Mstn protein expression was reduced significantly (P<0.001) with Sirt6 overexpression. H&E staining of the gastrocnemius muscles revealed that Sirt6 overexpression aided in maintaining the cross-sectional area of the muscle fibers. In our cachectic mouse model, we also observed that Sirt6 overexpression significantly downregulated (P<0.01) expression of CXCL10, an interferon-inducible circulating inflammatory chemokine highly implicated in many cardiovascular diseases.

Conclusion: Sirt6 could be a novel target for drug design. It would benefit patients by two ways: by impeding tumor growth and attenuating tumor-induced muscle degeneration.
