That in itself is a whole new subject............:)

Chuckie asked me abt CK1. The primary application for this is lung and pancreatic cancer in Trial 1 and thus my responses were specific towards his queries. Yes, there are other compounds and as said, Trial 2 is being used for other cancers with the compounds being used as a stand alone therapy without application of s-o-c chemotherapy. Can you elaborate pls WM...having difficulty deciphering today :)

everyone needs to understand is that in application towards lung and pancreatice cancer trials, CCT245737 is NOT a treatment per se. The standard of care chemotherapy is the primary treatment. But as per my posts labelled 1-4, due to the presence of checkpoints, the DNA damage which may be successfully treated by the chemo, can repair itself. The aplpication of CCT245737 is hoped to prevent the repair of the damaged DNA thus making the primary chemotherapy treatment more effective. Thus the two applications used in combination is hoped/expected to deal with a more permanent apoptosis of the mutated cells. Does that make sense? :)

Sorry WM, but you're mistaken on this point ''it does happen'') :) There are 2 phase 2 clinical trials being conducted. The trial using CCT245737 ultimately being used for targeting lung and pancreatic cancer patients, will be will be used in combination with standard-of-care chemotherapy, not as a stand alone therapy. The chemotherapy is required to initially target cancer cells but CCT245737 is then used for the checkpoints i.e in order to prevent the DNA damage from repairing itself following application of the standard of care chemotherapy. However, the second tiral will use CCT245737 as a single anti-cancer agent but will intiially target other/ various cancers. Make sense? :)

I’ve noted SAR have also state ‘’additionally, some cancers, particularly those where the cancer cells are undergoing 'replication stress' are expected to be treatable by a CHK1 therapeutic without the need for chemotherapy.’’ The fact it is being used in conjunction with ‘Standard of Care’ simply means it is used conjunction with an already established protocol for the specific cancer, lung and pancreatic in this case. This will be an established medical treatment dependnet on the type of patient, type of cancer and/or clinical conditions, pre-determined via extensive scientific research. ***The main point to take away is the ‘checkpoints’ and how SAR’s compounds disrupt them thus potentially preventing repair of DNA damage mutation).*** HTH? :)

CHK1 kinase has been shown in studies to play an important role in: a) DNA damage checkpoint control b) Embryonic development c) Tumour suppression SAR’s therapeutic compound, CCT245737, inhibits CHK1 kinase function. It is being targeted in treating proliferative diseases (e.g. cancer) mediated by CK1 that are then ‘’ameliorated by inhibition of CHK1 kinase function’’. Simple terms, the aim of CHK1 therapeutics is to ultimately cause disruption to the specific ‘checkpoints’ with the aim to prevent cancer cells from being able to repair the DNA damage, hence improving the extent of tumour cell death caused by chemotherapy.

Many cancer cells can repair this DNA damage, therefore making the chemotherapy less or ineffective, via what is known as ‘checkpoints’ that take effect during the development of the cancer cell. These checkpoints give a tumour cell the opportunity to repair damaged DNA caused by the chemotherapy thus making it less effective …and the cancer either not being completely eradicated or returning.

Cancer is essentially the mutation at specific points of the genetic code (DNA) within cells coupled with uncontrolled growth (oncogene genetic mutation). Chemotherapy normally takes effect via DNA damage of the cancer cells leading to cell death (apoptosis) and the tumour shrinking. It generally uses anti-cancer (cytotoxic) drugs to destroy cancer cells by ceasing or impairing reproduction (mitosis) leading to apoptosis of these fast diving cells. Mitosis is prevented via various mechanisms including DNA damage and cell division inhibition. Most target-specific therapies ‘attempt’ to target abnormal gene expression. The drugs (in theory) will specifically target only those areas of the DNA code that have ‘mutated’ and not the part of the DNA code which remains ‘normal’. However, in ‘practice’, normal/healthy cells are also affected thus leading to a whole multitude of varying side effects associated with different forms of cancer.

Had a quick look on SAR (you know why I keep away!). Saw these two posts of yours but they're kind of confusing re the questions you wanted answering. So I've formulated a response (apologies if any of it is too basic-not being patronising) but don't know extent of what you know and not really sure what you're asking.
