Unfortunately LD didn't say how many mice this was in, but I would assume to be able to draw conclusions they will have used a number that is appropriate for this experiment. The three R's of animal research (replacement, reduction and refinement) here will influence whether there is more work on the combo testing. I suspect that ethically this work may not be expanded much further due to this, though unsure as what the sample size for this was and so whether it needs expanding to give a firm answer.
If you look at the ****** meier chart you see the following:
Control
Day 22 - 70% survival
Day 29 - 20% survival
Day 55 - 10% survival continuing until Day 60 (end of graph)
PD1 only
Day 22 - 90% survival
Day 24 - 60% survival
Day 32 - 50% survival
Day 35 - 40% survival
Day 42 - 30% survival continuing until Day 60 (end of graph)
Modi-1 only
Day 22 - 90% survival
Day 29 - 80% survival
Day 38 - 60% survival continuing until Day 60 (end of graph).
Modi-1+PD1 combo
Day 42 - 90% survival continuing until Day 60 (end of graph)
Take home points - PD1 actually progresses relatively similarly to the control mice. Frustratingly there is no comparison between Modi1 alone and control, so we can't see whether this is statistically significant, but eyeballing the graph it looks as if it is (probably 0.01 I would guess). The fact that Modi-1+PD1 combo, vs Modi-1 alone is not significant is a plus as it suggests that Modi-1 is the driving force behind the survival of these animals. Hence actually improving this significance, would suggests synergy (i.e. Modi-1 not having as much of an effect), decreasing it would suggest that the Modi-1 combo is being driven more by the Modi-1 peptides rather than PD1 which could be seen as a benefit as a stand alone therapy. I guess this could be interpreted as a negative if SCLP were planning on licensing Modi-1 for use with PD1 combo therapy rather than outright sale. So depending on which direction the company want to take this in, depends I think on whether you want to have Modi-1 working better as a stand alone or a combo therapy.
