This seems like a sensible move. As Scancell grows a joint CEO and CSO role is too big for one person and much better that Lindy concentrates on driving the pipeline. Dr Phil L'Huillier appears to be an excellent appointment and he's certainly well connected with plenty of experience.

Scancell have traditionally always held their AGM in mid to late October. We've discussed the timing of the 2024 AGM here before and I believe someone contacted Scancell who confirmed that it was simply a reflection of the availability of the audited accounts. I've added to your list and as you can see it paints a slightly different picture.

Not sure how events that took place months ago can tell us anything about whether or not the 85% ORR is being maintained. We already know that this study has a very high probability of success and Scancell are simply planning in advance the next stage of development in order to move as rapidly as possible.

An ORR of 70% is required to meet the primary endpoint of this trial which would mean the trial had been successful. That would be fabulous and it's enough. If the ORR turns out to be 85% that would be a bonus, but it would be a shame IMO to raise expectations to expect this.

I wish I knew how they calculated mean PFS. As I say if it was median PFS then that would simply mean that at 10 months they still hadn't reached the stage where greater than half of the patients had progressed. I don't know, but would assume that before you can calculate mean PFS you'd need to know when all patients had progressed so perhaps they're simply saying that at 10 months in they can't calculate mean PFS because a/some patients still haven't progressed.

No doubt all will become clear when we get the much needed update for Modi1.

But what exactly does this sentence mean?:-

'In metastatic renal cancer patients mean PFS has not been reached at 10 months'

I'm not sure Johnny - I think they must surely be referring to survival of either the combination (single CPI) or monotherapy group, otherwise it's a pretty meaningless statement. They could simply say for every cohort for both trials that 'X' target hasn't been met yet. Whilst PFS is being measured, I'm not sure that there is a target suvival time period either.

I have no idea how they calculate mean progression free survival but if you substitute the word 'median' for the word 'mean' then it does make perfect sense so I suspect the answer lies in understanding exactly what they mean by 'mean PFS'.

Yes confusing to me too, I could understand if they were saying median PFS hadn't been reached at over 10 months. Also not sure whether they're referring to monotherapy or combination patients.

Definitely promotions of existing staff rather than new recruits and good to see Scancell supporting their career development.

ee - headcount increased from 51 to 61 during the last finanacial year, all 10 new recruits were R&D staff (43 to 53) with the admin and other staff headcount remaining unchanged at 8. So resources being focused where they're needed with a 25% increase in R&D staff.

Link to the project is below. This is much more about analysis of tumours, the tumour environment and immune responses to understand why some patients respond and some don't and why some experience really severe side effects whilst others taking the same therapy don't. So carrying out multiomic analysis including looking at areas like the microbiome to identify predictive biomarkers which may also potentially become targets for new therapy. There have been incredible advances in the tools available to carry out this analysis such as next generation sequencing etc. and hopefully the project will lead to greater understanding of how cancer and the immune system operates at a molecular level.

Of course big pharma's involvement isn't motivated by altruism and you're right to highlight this. The UK is a world leader when it comes to genomics and the NHS is ideal for studies like this that require many thousands of samples and that's why they want to be involved. It's not necessarily all about personalised treatment, big pharma also have pipelines stuffed full with other non-personalised therapies and if the project is successful we all (including Scancell) will benefit.

A bit of a negative take on an important project. Anything that advances our understanding as to why patients fail to respond to therapy or suffer severe side effects surely has to be a good thing. The potential benefits go beyond reducing unnecessary suffering and saving lives, if successful it could save healthcare systems huge amounts of costs and resources and streamline the whole drug development process.

Who knows, Scancell may even benefit from their findings but I think it's great to see the UK focus on such an important issue.

Don't think it's a case of the AGM being particularly early, just that it was late for the past couple of years. Am sure the timing has been driven by nothing other than the availability of the board, half of them aren't even based in the UK and all will have very full diaries.

The Texas Biomedical mAb was created by cloning antibodies from patients vaccinated with the Pfizer Covid vax and targets the spike protein of the SARS-CoV-2 virus. It's being developed as a broadly neutralising Covid therapy. Scancell's SC27 targets theLewisy glycan and is being developed as a therapy for gastric cancers.

I think that may be the case for the nivo element of the doublet therapy where patients with higher levels of PD-L1 expression are more likely to respond but I don't recall Scancell ever mentioning any biomarkers that are predictive of response to SCIB1.
