PFIZER ADVANCES BATTLE AGAINST COVID-19 ON MULTIPLE FRONTS

Ahfam from what I see TYK2 dosnt seam to be talked about as a solution for COVID-19 but more JAK related as the article suggest, if so then Pfizer would be in prime position with as Num4 suggested with (human safety data) with their JAK1/TYK2 inhibitor but then they are advertising their other selective JAK as a solution and not the JAK1/TYK2 inhibitor, so no clear evidence in my mind, the focus is Autoimmune and the first Cancer related selective Tyk2 that’s where it is at!

Looks to be the JAK associations in the latest research, 27th March 2020!

JAK inhibitors

JAK inhibitors work by inhibiting the activity of one or more of the JAK family of enzymes, including, JAK1, JAK2, JAK3 and TYK3. JAKs interact with signal transducer and activator of transcription proteins (STATs) and the JAK-STAT pathway is central to cellular response to exogenous signals in the immune system. The JAK family of enzymes are responsible for signal transduction and JAK inhibitors play a major role in inhibiting and blocking cytokine release that can contribute to growth of malignant cells in cancer. JAK inhibitors are used in the treatment of cancer and inflammatory diseases such as rheumatoid arthritis [43].

This review focuses on how the JAK-STAT pathway can be manipulated to reduce viral entry and inflammation in patients with coronavirus. The main targets that the review highlighted were baricitinib (JAK inhibitor), IRE1a (an endoplasmic reticulum stress sensor, leading to an increased expression of negative regulators of JAK-STAT SOCS1 and SOCS3) and combination therapies using tylophorine-based compounds with JAK2 inhibitors.

Conclusion

Current studies suggest that although there are potential targets in the JAK-STAT pathway that can be manipulated in the treatment for coronaviruses, they are all in early stages and require further in vitro and in vivo studies to confirm their therapeutic effects.

Pfizer have their own similar TYK2/JAK1 in Phase 3 study’s so I don’t think that’s right guys!

I’ve noted that this site is monitored and when research appears here I appears as a tweet to up sentiment, it’s a PR company doing imo!

The research has gone into fine tuning 2 kinase molecules into 2 types of Deceases Autoimmune and Cancer lagging behind, it would seam unusual to focus attention on covid19 as another lead molecule would have to be created as it’s taken years to get the molecules into pre-clinical which still is not completed, if they were going to focus energy into a pneumonia related molecule or speak of it too much as in “we are now focusing effort to enter this” then I think from my point of view they would loose credibility switching the business plan and taking focus away from what’s put in place already, they have acknowledged the facts based on the Tyk2 pneumonia article which is good enough for me, the action seams to be IL-6 related which to my previous knowledge hasn’t been mentioned before in relation to 1801 could be wrong as haven’t looked in depth but to my mind right now, and the IL-12 and IL-23 has been mentioned and I’m not taking away that there would not be a link with 1801 properties has in relation/relevance to covid19 and a more taylored molecule for this but right now we haven’t got anything in place for this imo!

CFP this has been said by Tim a lot over the years, but I do understand what your saying with Nimus pre-clinical program for all their Tyk2 compounds being licensed early but as this is what has been stated and the Patent separation issue then this is what I’m thinking!

CFP those hard decisions can’t be made until preclinical is done or nearly done (late preclinical too early Clinical) whose are the stated timelines which makes sense, and the patents are settled to conclusions, if we get any milestones the money in my mind with further the Tyk2 deeper into bigger deal territory, they will make the decision into what can be concluded given the cash flow my bet would be it’s a carbon copy of SRA737 get mhra approval and start the clinical trials of either SDC-1801 or 1802!

They are always talking to interested party’s as has been for a while and would bet a few as you would never just have one interested party, with the current cash then I hope they start clinical trials before getting a deal, most money to be had would have at the least get pre clinical done and complete on going long term toxicity study’s, I think judging by the presentation then end of the year, with IND application approval patents would be further along if not granted too!
