There has been an understandable trend in recent years to more sophisticated, more complex medications as the low hanging fruit has been picked and largely found to not be very satisfactory in reversing cancer. The modern treatments are VERY expensive. In this landscape, the pre|CISION technology is a no brainer as long as the prodrug - the drug+leaving group conjugate - is shown to be inert other than cleavage by FAP. Impossible to put a valuation on this but it will be very high.

FAP isn't just upregulated in tumours, it is also present in high numbers in other conditions. The AVA6000 actually lists the conditions:

"Disclosed are prodrugs of cytotoxic anthracyclines (such as doxorubicin) and other therapeutic agents that are selectively cleaved and activated by fibroblast activating protein (FAP). The prodrugs are useful for targeted delivery of cytotoxic and other agents to FAP- expressing tissues, including cancer (e.g., solid tumors). I'm not familiar with modes of action but if, say, you have a powerful rheumatoid arthritis drug that acts at the site of arthritic inflammation but has unacceptable side-effects elsewhere in the body, then that might be a suitable candidate to present as a prodrug using the pre|CISION platform.

and someone recently posted a link to a Fred Hutchinson call for patients that highlighted breast cancer. So it is my educated guess that Avacta and the clinicians want to cover all or as many of the susceptible cancers as they can in order to a) be able to demonstrate 'initial efficacy' in as wide a range of epithelial cancers as possible in Phase 1a, and b) speed up recruitment.

What he said was that it might go to C7 (which they currently have authority to do, up to 400mg/m²) but he couldn't see it going beyond. One has to trust that the clinicians have a feel for when MTD is being approached (white blood cell problems) so it's not AS nor Avacta who're deciding how safe and tolerated AVA6000 is.

Now, assuming you mean DE7 as in C7 and not C8 - AS has muddied the water by starting to refer to the cohorts as 5th dose escalation [cohort] and 6th dose escalation [cohort] - then C7 could be 370, C8 430 and C9 490mg/m². That last one would contain the equivalent of 330.75mg/m² doxorubicin. IN EACH DOSING CYCLE. Just let that sink in... Given that doxorubicin from AVA6000 has been found in (above) therapeutic levels as well as halting disease progression in some patients at the 160mg/m² dosing level, so from C3 onwards, C9 would be approximately TRIPLE that C3 dose and we'd be talking about tumour regression rather than just halting disease progression. And you think funding might be a problem?
