Indeed, the majority of KOLs stated that allosteric mechanisms are not unique and orthosteric TYK2 inhibitors could possibly achieve comparable levels of efficacy and safety.55 One KOL submitted that “there is no evidence that an allosteric action mechanism leads to any benefits for the patient (in terms of efficacy or safety). Nor is an allosteric mechanism necessary to target a TYK2”,56 while another KOL added: “increased selectivity […] does not necessarily characteri[s]e every inhibitor with an allosteric mechanism”.57 Similarly, a competitor who is currently developing a TYK2 inhibitor stated that “overall, the use of an allosteric or an orthosteric mechanism for TYK 2 inhibition does not make a difference for the patient (i.e. in terms of efficacy or safety). […][A]n allosteric mechanism for TYK 2 inhibition is not necessarily more targeted than an orthosteric mechanism and may give rise to similar side effects.”58 For this reason, the Commission concludes that, at this point in time and on the basis of the elements at its disposal, no separate relevant product market can be defined for allosteric TYK2 inhibitor treatments for moderate-to-severe PsO.

Further government funding for 1801 would need to be lined up before the end of the study to assure smooth progression, 3months of data I'm sure John knows enough to make an application to progress the compound, it took a very long time to make the 1st application, we can't have a 4month wait to progress if it is showing efficacy.
What progression looks like, in this world who knows, what it won't look like is a sub 2p SP.

Timelines they very recently committed to are coming to an end for a CTA on 1801 for a yet undisclosed autoimmune target. Tim must be sitting on plenty data at the minute.

It's interesting that Nimbus and BMS still haven't pushed their tyk2 into a COVID treatment scenario. Don't know why..
I wonder how much Sareums manufacturing route for tyk2 can produce as I can guarantee it won't be enough if it's a viable COVID treatment.

If they haven't finished all studies for 1801 by the half yearly then they are well behind on their timelines.
The CTA should be in by end of March.

Tim has the inside information which will allow him to hold the cards for any tyk2 deal, he will know how the COVID study is progressing thus ensuring a fully funded pathway to revenue.
To pry SDC1801 or SDC1802 from his hands your going to have to show a huge committment to the development, after all that is what he really wants. The billions and shareholder value are the byproduct of his success in helping treat life-changing disease.
The hint of positive lab results, which the science and other 1st gen drugs are giving us warm feelings about, means Sareum are the biggest under rated stock on AIM.

Yes Damion, I was looking into this today actually, we are awaiting results of their trial atm. Should be finished. Interestingly they did a food study.

I think it's only Rinvoq that doesn't have the warnings, to date.

They certainly don't feel confident putting their compounds into a COVID treatment trial, yet the research supports it. Why not...Too many side effects perhaps..

Hi Fadec, as I mentioned in the post - they have what I would term the 1st class inhibitors, all with black box warnings to date. If they believe in the class and want something cleaner then that's why I think they would look at Sareum. If nothing else to protect their pipeline, a competitor taking up these 2nd gen JAKi could impact their future sales. Sareum will have watched these molecules develop, the flaws and success - then designed accordingly. Hence 2nd generation JAKi.
