I think the main question they have for the FDA is to confirm that the trial can be run with the primary endpoint being improved stool consistency /bowel movement rather than that and pain relief.

This is because the FDA have previously said that but obviously before starting an expensive P3 trial they want confirmation.

The Blautix results were very good for stools and less so for pain.

It's for adjuvants to mucosal (nasal) vaccines.

I suspect they are for dendritic cell vaccine adjuvants for cancer as Merck have a couple of these in early clinical trials. I also suspect 0518 is involved as one of the candidates.

The agreement was for 3 years and is due to expire in October. It could of course be extended and that would likely need to involve a milestone payment as 4D can't do any vax stuff with other companies whilst working with Merck.

I wouldn't get too carried away with the $1bn total milestones. That would involve all 3 LBP candidates achieving approval and sales.

Merck invested in 4D as part of the deal and the 1st payment was upfront for $5m.

Other than that it's all hush hush.

We know that in addition to 20 renal patients 27 others were dosed in NSCLC, bladder cancer and a couple of other indications.

If they announce 1 in 10 from NSCLC then they could get another $7.5m but they still have to hold $7.5m in the bank without spending from the 1st drawdown.

FDA have delays all over the shop. 4D could have already had the feedback as receiving feedback would not be RNSable in itself. The strategy update is what matters.

The "antibodies" programme is presumably the nasal Vaccines collaboration. It is at an early stage - still pre clinical. It expires in October so will have to find out something by then.

I would love a good positive RNS in the morning that causes a decent rise as I still have a holding here.

I was practically all in and I am sure anyone can agree that is an insane way to run an investment portfolio.

I have now diversified to include some less risky stocks and even a couple of funds.

It seems quite likely that the company expected to be able to raise on the back of 0518 data release.

Hence the shelf reg filing.

The hope for a rise to make a fundraise possible lies with us lot and most of us don't want one. (I do if no other funding options are available).

I am just presenting my most up to date thoughts on 4D given that new info has come to light from the annual report.

In February I was still able to dream that 0518 would deliver a knockout update.

Instead we got an update on 20 patients dosed on the renal cancer arm. 4 out of 16 stable disease plus 4 dropouts.

Just 47 patients dosed in the whole trial out of 130. It was 24 dosed a year earlier so big recruitment slowdown with no explanation and no news on any of the other arms of the trial.

I was hoping for more. If I had been so sure it was below par I would have sold into the rise.

However, I wasn't and I was fooled by the initial positive market reaction.

I am now concerned that the delay getting the Bavencio trial going is funding related. It should have started towards the end of last year.

I really hope I am wrong. I had been over exuberant in my 4D buys and selling at a substantial loss was a consequence.

The trial results have introduced more uncertainty in terms of a potential Merck deal.

I actually was having suicidal thoughts last week when the price was dropping to 36p when I had bought more that morning at 40p.
