Surely we can all agree that Stay Long's comments are pointless and none of his comments are actual criticism of 4D which would require a rebuttal so it seems the only sensible thing to do is make him a little green box

This article from a few months back seems very important for 4D.

It is about the need to overcome Treg cells resistance to immune checkpoint inhibitors like Keytruda.

Slide 12 on the recent investor presentation is titled Overcoming Treg Resistance to ICIs with 0518.

The article suggests chemotherapy and some high tox low efficacy monoclonal antibodies as ways to overcome Treg proliferation.

I imagine most patients and doctors would prefer a safe and efficacious treatment like 0518 than chemo.

Yeah that's what I was think Tutto. I mean what would adding say $1bn a year (conservative estimate) in sales to Keytruda to Merck be worth?

Their price sales ratio is 4-1 so that would be $4bn in itself and doesn't factor in the value of the MRX drug discovery platform and the other assets.

They can also typically go straight into Phase 2 study. Blautix didn't have a Phase 1 that would have involved dose escalation etc.

It was straight into dosing IBS patients with pre defined optimal dose.

Same with the upcoming Bavencio trial in bladder cancer

I got a bit worried when I first read about that but e gallinarum strains are lower risk than other e strains like e coli

The assumption is that the single strain approach is inherently safe with low risk of infection due to the 4D production process.

There's a very small chance of having a strain like e gallinarum which is 0518 causing antibiotic resistant infection or possibly auto immune disease but they spend a lot of time when doing the strain selection and optimization to adopt safeguards against anything like that

Yep urgent unmet need with a therapy with little or no tox issues.

Of course if they announce 1 in 5 success in oncology (enough to get approval as Mercks original denominator of success was 1 in 10) we all know how the derampers and dumpers will play that as failure when in fact it would be great news.

It's 142 patients. 12 were dosed in the first part and there are currently 4 groups of 30 in 4 indications and they added another 10 for another indication (possibly breast cancer without checking).

They have said recruitment expected to be ongoing until the end of the year. Hopefully they will have accumulated data in say 50 patients or so in terms of the clinical update.

Of these if anything above 1 in 5 were responders it would be enough for future approval. All the more so if the responders can be shown to have the Treg ICI resistant biomarkers.

I would hope to see 2 in 5 responders for it to be absolutely mind blowing. Either way if they have the responders/biomarker data I would expect a P3 in this group to be started fairly quickly. (I am not sure how many patients would need to have data for conditional approval to be granted. The whole time would be subject to buyout)

Unfortunately it looks like it will be a struggle to provide the readout this side of Xmas but fingers crossed.

The pancreatic cancer will provide more safety data on 0518 added to Keytruda and the monotherapy trial. I haven't really investigated this enough as it is so likely to be dismissed by the market as only 15 patients but I will be very interested as it is yet another setting for 0518 and could potentially be combined with other treatments in addition to radiation.

Asthma is also very important. It is the asset the 4D scientists were most excited about before they got results back from 0518 lab work. I feel fairly confident they will be good and also good to see placebo used

Stay Long I think you were a bit silly to sell up but you obviously think you have found a better opportunity.

You are in up to your eyeballs yet you are not prepared to offer an opinion as per your expectations for the forthcoming trial readouts.

I don't know if you are aware but with a biotech clinical readouts and their interpretation by the market are really important.

So as readouts are due what would you hope to see if RNS with results came out tomorrow?

Looks like a pump and dump operation to me. Like what they did here.

At least 4D get that to get a drug approved you have to run trials, rather than boast about how good your product is without any evidence.

I read an interesting article the other day about red flags in pharma companies. It said watch out for biotechs that employ a scientist at the end of their career to provide positive updates and gravitas.

Made me think of Howard Weiner.

Dave it's very simple. If I make a comment on the price movement I am not running an orchestrated pump and dump in cahoots with others to run the price up, so that others buy in before dumping it and leaving them to hold the bag.

A lot of people here are only here because they got caught out by the last operation and so they are only too happy to support you in your endeavour so they can dump as well once it's at 110.

I also think it's a question of manners. Where else but on LSE would a bunch of strangers turn up acting like they own the place and insulting people that have been there much longer and will be there long after you have gone with your winnings to find your next victims.

That's a commentary on price movement not the company or it's prospects.
