GSK are achieving commercialisation for Dorstalimab in areas that not only our 737 were achieving great results in but also other CHK1 inhibitors.

737 halted development for several years that GSK fast tracked Jemperli as a treatment in the EU. Same fields of oncology as Chk1 inhibitors

Now we have 737 on hold for over a year and now have approval for endometrial cancer.

My opinion for what it is worth is that the likes of Jemperli face the threat of hefty competition from CHK1 inhibitors.

A question with the monoclonal antibody which recognises and can destroy the identified cancer cells, is how is the DNA damage repair pathway addressed?

Eventually treatment resistance will develop.

GSK have paid billions in out of court settlements involving marketing of their drugs this includes not giving the true picture.

737 sits on the back burner now.

We have absolutely no say or or influence over decisions or marketing of 737 at all.

All we have is a vested 27.5% financial interest.

CPF will do whatever CPF want. We have no influence.

We now sit with our 1801 and 1802.

As for just picking up the phone and making a deal, that just does not happen in the pharmaceutical world.

Triparna Sen pointed out the startling effect that PD-1 Chk1 and LDG combo.

Much work was done by Michelle Garrett with investigational studies with incombo in this field and gave detailed info in the reports with effect to efficacy DDR and resistance to treatments and indeed pointed out combinations whilst effective needed further investigations into safety.

Can the doubters explain their opinions as to Chk1i addressing areas of unmet need in its potential ability to treat cancers.

Why is Acrivon developing prexasertib and is fast tracked

I have been trying to buy fome more but not able to. That very odd.

Should SDC1801 reach its potential on best safety profile and phase2 ready you will be looking at 500 million plus or that minimum add in completion of SDC 1802 preclinical and that 250 million alone, then look at 737 bearing in mind we would need to know what develops are happening and in what indications a further 50 million.

So my guesstimate is 800 million minimum for lock stock and barrel.

Deucravacitinib turnover 137 million and forecast 2 billion turnover by 2030. That has failed in UC not overly efficient in Psoriasis but has best safety profile as hypothesised as an allosteric inhibitor utilising The Jh2 domain that supposedly regulates the JH1 domain

It is all about efficacy and having a very favourable safety profile.

Molecules used?

Ours are patented. Ot is important how they work and how they bind plus purity of the end product compound.

On receipt of continuing developments should put approx 10% of our share of 737 ie 10% of circa 80 million that being 8 million or 6 to 7p per share minimum.

People will sell some l believe should this be the case as we have a few months to wait on toxicology studies but then again that does not remove the possibility of licence or buyout in the meantime.

We are for what we have extremely undervalued and we are not the only ones undervalued on AIM.

The intention is to provide a list of current faults and see how we compare so far.

One important aspect is the increase in creatinine levels which may be indicative of renal damage can run from acute treatable through to severe but difficult to treat maybe leading to dialysis and of course keep going and renal failure as I death.

We have no observed no increase in creatinine levels so from that aspect extremely reassuring.

It is a competitive market and sooner or later these compounds will be superceded by better compounds of which ours should be one.

Breprocitinib and Ropsacitinib by Pfizer/priovant have been discontinued in most of their originally intended Indications.

There are reports in some trial results of increased creatinine levels above the >2x threshold and all they report is no incidence of this increase resulting in a death. Rather like running backward and forwards across a busy road and saying I have not been killed yet. Benn hit a few times haha

This is where careful interpretation of data is relevant.

They also state.'gave an acceptable safety profile'

Wow so acceptable that they decide against continuation in that indication.

These compounds whilst most likely capable will not be near the tip of the tree, therefore less in demand and blow me down the cost of treatment would have to drop considerably to remain competitive. Major loss of income.

Hence let's go for an indication into areas of urgent high unmet need.

Pfizer which is partially owned by Blackroch have put one if the compounds into HI or is it IH? Bu99ered if l can remember. Cancer also used as an indication to repupose into as for obvious reasons

long term safety not as important as I immunotherapy.

You only given 6 months to live you are not going to be concerned of a low risk 5 years down the road.

All about establishing the safety of the compound over longer term and proving via investigations that we are inherently safer than vo.pounds currently approved and nearing late stage clinical trial completion.

Breprocitinib and Ropsacitinib not performed as expected so a letdown for them there.
