JT referred to the biopsies which are in the science day deck and are pretty unequivocal, target concentrations at the bottom.

JT you said Ok great, but if ava6k was cleaved at too low a rate in the tumours, it is possible that there wouldn't be enough dox around to kill the tumour.

But surely the biopsies showed the cleavage working well as the ratios of dox to tumour versus plasma illustrated this and there a correlation between AVA6000 dose increase and increase in dox in the tumour.

The heavily treated patient population means signs of efficacy were meant to be highly unlikely. Main objective is of course safety and Avacta has asked for biopsies to show mechanism of action as although PK data will show this biopsies are unequivocal. The biopsies showed the drug is performing as it did preclinical and delivering a lot more dox to TME and sparing healthy tissues (plasma samples as well as no DLTs). Dox well understood so getting the volumes of dox into tumour will do what is has done for 40 years, the molecule won't suddenly change. We also have several patients still receiving drug as their disease is stable which is something in these heavily treated patients and we remarkably have significant tumour shrinkage. None of that was expected at this stage with these heavily treated, end of the road patients. Data read out will no doubt shed a lot of light and hopefully the comms teams have a well thought out strategy to execute.
